A case study of disseminated histoplasmosis linked to common variable immunodeficiency BJID
Rachid,Acir; Rezende,Lucila Stange; Moura,Suzelle Freitas de; Loffy,Paulo C.; Magalhães,Francisco Luiz Gomide M..
Histoplasma capsulatum, a ground fungus, can infect humans, normally in endemic areas; the resulting disease can be asymptomatic or it can have a benign development, but in rare cases it can develop into a serious clinical condition and can even be fatal. Its most characteristic initial location is in the lungs, resembling tuberculosis, often accompanied by mediastinitis and an exuberant fibrotic response. The spread of this infection can be caused by the concomitance of another illness that alters the immunological balance. Sometimes such an association is not clear. Therefore, disseminated histoplasmosis is defined as a clinical condition where the fungus is present in more than one location. Common variable immunodeficiency is characterized by a...

A new methodology for polyvalent intravenous immunoglobulin solution production with a two-stage process of viral inactivation BJPS
Lucena,Antônio Edson de Souza; Sampaio,Divaldo de Almeida; Silva,Ednaldo Rosas da; Paiva,Virgínia Florêncio de; Santiago,Ana Cláudia; Leite,Ana Cristina Lima.
Highly purified intravenous immunoglobulin G concentrate (IV IgG) was produced with the use of polyethylene glycol associated to a single-stage precipitation by ethanol, instead of the classic Cohn-Oncley process, which employs cold alcohol as the precipitating agent, in a three-stage process. Precipitation of crude fraction containing more than 95% of immunoglobulin G was performed by liquid chromatography with a cation exchanger, CM-Sepharose, as a stationary phase. During the process, the product was subjected to two-stage viral inactivation. The first stage was performed by the action of sodium caprylate, 30 mM at pH 5.1+/- 0.1, and the second stage was performed by the action of a solvent-detergent mixture. The finished product was formulated at 5%...

Antibody and cytokine serum levels in patients subjected to anti-rabies prophylaxis with serum-vaccination J. Venom. Anim. Toxins incl. Trop. Dis.
Ayres,J. A.; Barraviera,B.; Calvi,S. A.; Carvalho,N. R.; Peraçoli,M. T. S..
Rabies is considered a fatal disease once clinical symptoms have developed. The aim of this study was to evaluate epidemiological aspects and immune response in patients attacked by domestic and wild animals and subjected to post-exposure rabies treatment with equine serum and associated vaccine. Thirty-three patients were evaluated; they were between 13 and 65 years old, 75.8% were male and 24.2% female, and from the Botucatu neighborhood. Twenty healthy control individuals with the same age range were also studied. Specific antibodies to equine immunoglobulins and IFN-gamma, IL-2, IL-4, and IL-10 production were evaluated by ELISA. IgM, IgE, IgG and subclasses, and rabies virus antibodies serum levels were determined by nephelometry and...

Economic evaluation of snake antivenom production in the public system J. Venom. Anim. Toxins incl. Trop. Dis.
Morais,V.; Massaldi,H..
Snake antivenom, an expensive animal product, is presently the only effective treatment for the consequences of snakebite. In Latin America, antivenoms are mainly produced by public institutions with frequent shortages of the necessary supply. Here, we present an economical analysis of the factors affecting production cost, assuming a basic processing batch of 100 L hyperimmune plasma. Three annual production volumes were considered for two typical production technologies. The components of cost were classified as fixed, variable and semi-variable. We found that in all stages of production, fixed cost represents the major contribution to total cost, and is given essentially by manpower cost, particularly for low production volumes. Our estimation shows...

Effect of Embryo Thermal Stimulation on the Serum Levels of Immunoglobulins and Corticosterone, and Organ Histopathology of 1 day-old Broilers Rev. Bras. Ciênc. Avic.
Flores,F; Nääs,IA; Garcia,RG; Quinteiro,WMQ; Souza,LI.
ABSTRACT Embryo thermal stimulation has been studied as a means to promote epigenetic changes and to improve broiler health and performance. This study aimed at evaluating immunoglobulin (IgM, IgG, and IgA) titers, serum corticosterone levels, and organ integrity of day-old male Cobb(r) and Ross(r) broilers. Embryos were submitted to hot and cold stimuli (thermal stimulation) on days 14-18 of incubation. The thermal manipulation treatments did not affect immunoglobulin titers of 1 day-old Cobb(r) or Ross(r) chicks, and caused severe bursal lesions (scores 3 and 4) in both strains. However, serum corticosterone levels of Cobb(r) chicks were higher when embryos were submitted to hot stimulus (+1.39ºC) compared with cold stimulus (+36 ºC). Serum...

Effects of Methionine Deficiency on the B Lymphocyte and Immunoglobulins of Cecal Tonsil in Cobb Broilers Rev. Bras. Ciênc. Avic.
Zhu,M; Ruan,T; Zeng,Q; Wu,B.
ABSTRACT The present study was conducted on one-day-old broilers to investigate the effects of methionine deficiency on B lymphocytes and immunoglobulins (sIgA, IgA, IgG, IgM) in the cecal tonsil of Cobb broiler chicken. Methods including immunohistochemistry (IHC), western blot (WB), quantitative real-time reverse transcriptase-polymerase chain reaction (qRT-PCR) and Enzyme-Linked Immuno Sorbent Assay (ELISA) were used, and the study lasted for 6 weeks. It was found that the IgA+ B lymphocytes mainly existed in the diffuse lymphoid tissues and lymphoid follicles of the cecal tonsil. The data indicated that the number of IgA+ B lymphocytes was decreased in the methionine deficiency group, and the WB and qRT-PCR results suggested that the protein and mRNA...

Identification of enteropathogens from buffalo calves with and without diarrhoea in the Ribeira Valley, State of São Paulo, Brazil Braz. J. Vet. Res. Anim. Sci.
RIBEIRO,Márcio Garcia; LANGONI,Hélio; JEREZ,José Antônio; LEITE,Domingos da Silva; FERREIRA,Fernando; GENNARI,Solange Maria.
Faecal samples of 106 buffalo calves aged 3 to 45 days were collected sequentially, once a week, during six weeks for parasitologic examination. For bacteriologic and virologic exams, faecal samples were collected from all diarrhoeic and the same amount on non-diarrhoeic calves. Blood samples from calves were collected at 3 to 10 days of age for serum IgG determination. Eimeria spp, Strongyloides papillosus and Toxocara vitulorum were the most common parasites. E. coli, Enterobacter cloacae and Klebsiella pneumoniae were the enteropathogens most frequently found (alone or in association with other microrganisms). Heat-stable (STa) enterotoxin was detected in 5 out of 34 samples of E. coli isolated from diarrhoeic calves. Among the antimicrobials tested...

Immunoglobulin production is impaired in protein-deprived mice and can be restored by dietary protein supplementation BJMBR
Amaral,J.F.; Foschetti,D.A.; Assis,F.A.; Menezes,J.S.; Vaz,N.M.; Faria,A.M.C..
Most contacts with food protein and microbiota antigens occur at the level of the gut mucosa. In animal models where this natural stimulation is absent, such as germ-free and antigen-free mice, the gut-associated lymphoid tissue (GALT) and systemic immunological activities are underdeveloped. We have shown that food proteins play a critical role in the full development of the immune system. C57BL/6 mice weaned to a diet in which intact proteins are replaced by equivalent amounts of amino acids (Aa diet) have a poorly developed GALT as well as low levels of serum immunoglobulins (total Ig, IgG, and IgA, but not IgM). In the present study, we evaluated whether the introduction of a protein-containing diet in 10 adult Aa-fed C57BL/6 mice could restore their...

Renal involvement in canine leishmaniasis: a morphological and immunohistochemical study Arq. Bras. Med. Vet. Zootec.
Soares,M.J.V.; Moraes,J.R.E.; Moraes,F.R..
IgG and IgM deposits in kidneys of dogs with visceral leishmaniasis (VL) were studied in 25 symptomatic dogs (case) and 15 asymptomatic dogs (control) by an immunohistochemical method. All tested dogs were positive for VL by polymerase chain reaction, enzyme-linked immunosorbent assay, and indirect immunofluorescence test. Kidney fragments were submitted to immunohistochemical reaction. Many morphological patterns of distribution of subendothelial granules were identified for IgG and IgM in glomerular capillaries: global, segmental, diffuse, or focal. Intensity of immunohistochemical reaction to IgG was not significantly different when comparing the symptomatic and the asymptomatic animal groups by Fisher's exact test. IgM reactions were significantly...
